Importantly, another bottleneck at 13 days post-infection was observed, suggesting another bottleneck or selection process. Moreover, our study suggests that the selection occurred at the amino acid position 136, which is a residue located in the viral neuraminidase protein. Large particulate matter (PM) air pollution is a major public health concern worldwide. Yet ultrafine particles (UFP), defined as particles with diameter smaller than 100 nm, are much smaller and thus expected to be more toxic than PM10 or PM2.5. Despite growing interest, there are currently no methods available to directly evaluate UFP toxicity. Herein, we developed an in vitro human neuroblastoma cell line (SHSY-5Y) model and assessed the toxicity of UFP in three different modes: cytotoxicity, oxidative stress, and apoptosis. We determined that the SHSY-5Y cells were viable up to 24 h after exposure to two different commercial grade UFP particles. By contrast, we found statistically significant increases in oxidative stress after 24 and 48 h of exposure to UFP, consistent with dose-dependent cell death in all three UFP groups. Surprisingly, we also observed a statistically significant dose-dependent increase in apoptosis (up to 24 h) only in the two highest UFP dose groups. Our cell model can serve as a useful predictive tool to assess the toxicity of UFP in future studies.Q:
